Conditions: Column conditioning:
Convert column to the NH4+ form by rinsing with 10 ml 1 % (w/v) ammonium acetate
solution, followed by 10 ml dist. water. Slowly percolate 2 ml of a 1 % (w/v)
silver nitrate solution into the column. Rinse with 10 ml methanol, 10 ml
dichloromethane and 10 ml n-hexane.
Sample application:
Force or aspirate 1 ml sample solution through the column
Column washing: 2 ml n-hexane
Elution:
Separation of the fatty acid methyl esters acc. to carbon number and number of
double bonds (x : y)
Elute the conditioned column with 2 ml each of the sequence of eluents given
below. The amount and type of the fatty acid methyl esters is determined by GC.
Composition of the eluents (v/v) A to K:
A) 1 % dichloromethane in n-hexane
B) 5 % dichloromethane in n-hexane
C) 10 % dichloromethane in n-hexane
D) 50 % dichloromethane in n-hexane
E) 100 % dichloromethane
F) 3 % methanol in n-hexane
G) 10 % methanol in n-hexane
H) 50 % methanol in n-hexane (lower phase)
I) 100 % methanol
J) 1 % acetonitrile in methanol
K) 10 % acetonitrile in methanol
